Cannabis has been studied to obtain asexually propagated plants through tissue culture.
Cannabis genotype research
The selection of genotypes is very important for the success of tissue culture. Different species and different genotypes of the same species often differ dramatically in their morphogenetic capacity. However, it is worth noting that the more genetically and related the culture materials are, the more similar are the conditions required for morphogenesis.Feeney M. et al. applied four different Cannabis genotypes UnikoB, Kompolti, Anka, and Felina34 for tissue culture and Agrobacterium-mediated.SlusarkiewiczJaraina A. et al. used five cannabis varieties (Silesia, Fibrimon24, Novosadska, Juso15 and Fedrina74 as test materials for healing tissue induction and plant regeneration, and among all types of exudates the differentiation rate of the cannabis variety Fibrimon24 reached 61.92%.
Cannabis seed sterilization study
The choice of seed disinfectant and the determination of disinfection time are the key to effective tissue culture. Liu Yifu et al. soaked cannabis seeds in water with a concentration of 0.1% ascending mercury for 30 min, inoculated onto agar medium, and germinated to obtain sterile seedlings. Zhang Liguo et al. used a concentration of 10% sodium hypochlorite as a disinfectant for three treatments with disinfection times of 18, 21 and 24 min, and the germination rate was highest after 18 min of disinfection with a concentration of 10% sodium hypochlorite without causing contamination.
Physiological age study of Cannabis sativa
The time of taking the explants is also a key factor affecting the success of histoculture, and in general, the seedlings have better viability and regeneration ability. Liu Yifu et al. used the middle and upper leaf stems of Cannabis sativa hybrid generation and seedlings of good varieties (lines) with true leaves grown to 1 to 2 pairs as explants. Yin Pinxun et al. took the stem tips of the seedlings when they grew to 5-10 cm in height and treated them.
Studies on the origin of the explants
RichezDumanois C. et al. showed that the selection of explants is very important for the 2 cannabis plants for fiber. In the reported literature related to cannabis tissue culture, the main exosomes selected were: leaves, hypocotyls and stem tips. Zhang Liguo et al. selected the hypocotyls of the new cannabis variety Dragon Cannabis I for tissue culture. Yin P.S. et al. used the nodulated stem segments of industrial hemp variety Yunma No.1 as explants for in vitro culture and rapid propagation studies.Loh W.H.T. et al. used the stems and leaves of Cannabis sativa L. for the induction of healing tissues.
